Effects of rmr1 gene complementing synthetic lethality with nab2 knockdown allele on mRNA export in fission yeast

Abstract
We previously isolated of three mutants (SLN1~3) that showedsynthetic lethality in combination with the repressible nab2 allele,to identify genes involved in mRNA biogenesis and export infission yeast, Schizosaccharomyces pombe. In this study, uap56,rmn1, and SPCC1442.04c genes that complemented the growthdefect of SLN2 mutant were isolated. The SPCC1442.04cgene encodes a protein that is homologous to Saccharomycescerevisiae Rmr1 involved in gene conversion during meiosis. Thermr1 (SPCC1442.04c) null mutant showed normal growth andmRNA export. However, its overexpression caused deleteriouseffect on growth accompanied by intense accumulation ofpoly(A)+ RNA in the nucleus. Rmr1-GFP protein is localizedpredominantly in the nucleus. In addition, Rmr1p was coimmunoprecipitatedwith Rmn1p, which is a subunit of NURS(nuclear RNA silencing) complex involved in RNA silencingof meiotic gene expression. These results suggest that the S.pombe rmr1 (SPCC1442.04c) may have functions in linkingmRNA biogenesis to mRNA export and RNA silencing.
